Studies on in-vitro rooting and hardening in ginger (Zingiber officinale Rosco)

Abstract:

The present experiment was carried out with the aim to Studies on In-vitro Rooting and Hardening in Ginger of variety Mahim (local) using different rooting and hardening medias in CRD experimental design. The study was carried out at plant tissue culture laboratory of Dr PDKV, Akola during year 2023-24 in two phases with the aim of standardizing media for in-vitro rooting of regenerated shoots of ginger and to standardize hardening protocol for ginger. The shootlets generated in the in-vitro culture were placed in different rooting medias, where Half B5 medium in combination with 1 mg/l NAA and 1 mg/l IBA demonstrated a stronger response than the other medium. Once the culture had sufficiently developed its shoots and roots, the healthy in-vitro regenerated plantlets with a good number of roots were carefully removed from the culture jar, washed and treated with fungicide and were placed in portrays packed with different types of hardening medias. Out of all the hardening treatments tested, the ex-vitro acclimatization of plantlets yielded the best results in 100 percent peatmoss media with 100% survival rate.
